It Takes 7 hours and 20 minutes to drive from Orlando to Key West. That of course has many variables including traffic and the amount of times you stop. We highly suggest you do not take US-27 as it takes by far the longest.
Take the turnpike from Orlando to Miami and then to Key West. Taking I-4 to Tampa and then heading down I-75 may be a little more scenic but it takes an extra hour and makes you more likely to get delayed.
